Central effects of some synthetic quinazolones

The central effects of 5 newly synthesized quinazolones were studied. Gross behavioral studies in rabbit and mice showed that only dimethylquinazolone, like methaqualone, could produce ataxia and hind limb paralysis of longer duration suggestive of a central muscle relaxant activity. Higher doses, abolished straub's tail response, suggesting a peripheral muscle relaxant component. Pentobarbitone potentiation and SKF 525-A [2-(diethylamino)ethyl-2,2-diphenylvalerate hydrochloride] pretreatment studies revealed that dimethylquinazolone is a much less potent hypnotic than methaqualone whereas other quinazolones did not exhibit significant muscle relaxant activity.
